[...] Thus the study allows the molecular identification of tiger (_P.t.t_) and assigning their gender simultaneously by adopting a new approach of primer designing known as Allele specific- ARMS PCR technique. This technique allows the identification of unique nucleotides on the tiger target mitochondrial Cytochrome B (_mtcyB_) sequence and is upheld for designing tiger specific primers with a purposeful mismatch base insertion at their penultimate 3' end thereby making the markers much more specific in attending the assignation. The technique imbibed in three modes of designing primers for tiger and leopard for identifying fresh, decayed tiger samples and distinguishing leopard origin ones from that. The first mode of tiger designed primers (TF/TR) is an unlabeled and highly answerable with the fresh and also with the samples on a long stay of 20 days producing its identical amplicon with easily resolving length. While the second mode of tiger primers (TSF/TSR) is FAM labeled one which clarifies with the tiger samples by producing fluorescence on detection of the unique tiger base profoundly out of which the primers were designed. The assay produces the amplification with the short fragments which is cautiously resolved during the electrophoresis and showed an optimal amplification plot without any uncertainties. Third mode identifies its sympatric species leopard (_P.p_) from the samples as an alternative screening of its presence. [...] |
