Metode Non-invasif untuk Menentukan Pedigri Harimau Sumatera (_Panthera tigris sumatrae_) berdasarkan Penande D-loop dan Mikrosatelit

Sumatran tiger (_Panthera tigris sumatrae_) is the only surviving Indonesian tiger. It is an extinct species and categorized as critically endangered one. Analysis on genetic characters is able to design program in preventing extinction. This study was aimed at analyzing pedigree of Sumatran tiger based on genetic characters using control region HVS-I and microsatellite markers. DNA was extracted from hair of Sumatran tiger, consisting of one family. Hair samples were collected from Bandung Zoological Garden. Intact DNA samples were then apmlied using method of Polymerase Chain Reaction (PCR), prior to sequencing. Nucleotide sequence of control region marker was analyzed using MEGA IV software. It was compared to Aceh's Sumatran tiger (A1) using multiple alignment. There were 208 nucleotide differences between study samples (M1, W2, and Y4) and A1. Amplification product of microsatellite loci FCA 96 was used as a marker to assess individual genealogical relationship. Amplification product of the three study samples signified equal and parallel bands, therefore, it can be concluded that those samples were originated from the same pedigree, which was proved that the cub's heterozygote alleles inherited from both parents.
